The Spanish combination, known as Nadalcaraz, enthralled a packed and noisy crowd in a hot Court Suzanne Lenglen venue as they blended their considerable firepower to keep their gold medal quest alive.
Alcaraz, 21, smashed a forehand winner to earn the critical break of serve in the first set, with Nadal shouting his appreciation.
Koolhof, a former doubles world number one, and Griekspoor battled hard in the second set before dominating a tiebreak to win 7-2.
